چکیده

Phytases are the largest group of feed enzymes increasing accessibility organic phosphorus for animals. Feed phytases usually sold as dried powder secreting producers, mainly micellar fungi. We proposed a new technology producing phytase from Obesumbacterium proteus (OPP) in yeast Yarrowia lipolytica cytosolic protein (encapsulated OPP), where capsule (yeast cell) protects enzyme unfavourable factors (acid medium and active proteolysis stomachs) releases it along with substrate duodenum only. Here we report results testing encapsulated OPP on model broiler chicken comparison to conventional Aspergillus ficuum. The at dosage 30 FYT/kg provided maximum body weight end experiment equal or somewhat higher than control group, available deficit was complemented mineral supply. In contrast, soluble 100 1000 not able compensate diet, although chemical analysis demonstrated much diet non-accessible form. decreased residual Pi faeces by 2.1 times when added whereas negligibly affected this parameter regardless dosage.

Improving the efficiency of homologous recombination by chemical and biological approaches in Yarrowia lipolytica

Gene targeting is a challenge in Yarrowia lipolytica (Y. lipolytica) where non-homologous end-joining (NHEJ) is predominant over homologous recombination (HR). To improve the frequency and efficiency of HR in Y. lipolytica, the ku70 gene responsible for a double stand break (DSB) repair in the NHEJ pathway was disrupted, and the cell cycle was synchronized to the S-phase with hydroxyurea, respe...

Brown pigments produced by Yarrowia lipolytica result from extracellular accumulation of homogentisic acid.

Yarrowia lipolytica produces brown extracellular pigments that correlate with tyrosine catabolism. During tyrosine depletion, the yeast accumulated homogentisic acid, p-hydroxyphenylethanol, and p-hydroxyphenylacetic acid in the medium. Homogentisic acid accumulated under all aeration conditions tested, but its concentration decreased as aeration decreased. With moderate aeration, equimolar con...

Engineering of a high lipid producing Yarrowia lipolytica strain.

BACKGROUND Microbial lipids are produced by many oleaginous organisms including the well-characterized yeast Yarrowia lipolytica, which can be engineered for increased lipid yield by up-regulation of the lipid biosynthetic pathway and down-regulation or deletion of competing pathways. RESULTS We describe a strain engineering strategy centered on diacylglycerol acyltransferase (DGA) gene overe...

High-throughput transformation method for Yarrowia lipolytica mutant library screening.

As a microorganism of major biotechnological importance, the oleaginous yeast Yarrowia lipolytica is subjected to intensive genetic engineering and functional genomic analysis. Future advancements in this area, however, require a system that will generate a large collection of mutants for high-throughput screening. Here, we report a rapid and efficient method for high-throughput transformation ...
